HEIDELBERG, Germany, May 4, 2023/PRNewswire/ — The European Wellness Academy (EWA), a wholly-owned subsidiary of the European Wellness Biomedical Group (EW Group), has signed a cooperation agreement with the University of Heidelberg on of medical research. The research project, titled “A Tendon Specific Double Reporter Transgenic Mouse Enable Tracking Cell Lineage and Functions Alteration in-vitro and in-vivo”, is funded through the EWA.
This research has developed and defined a line of transgenic mice with tendon-specific expression of a fluorescent double reporter system, which will satisfy an identified need for animal models that allow real-time monitoring of cell behaviors throughout development, growth, and tendon repair in vitro and in vivo.
The Scleraxis (Scx) promoter activates the mScarlet red fluorescent protein to detect lineage changes. The proximal promoter of type I collagen alpha 1 chain (Col1a1) controls blue fluorescent protein. Two promoter regions called tendon-specific cis elements (TSE1 and TSE2) ensure that blue fluorescent protein (BFP) is uniquely expressed in tendon tissues.
Based on these findings, the tendon-specific dual reporter system may be a unique paradigm that can be used to investigate cell redifferentiation as well as extracellular matrix modifications both in vitro and in vivo. The Scx-mScarlet transgenic mouse has stable fluorescent expression, making it an excellent tool for monitoring Scx cells during tendon formation and growth.
The objectives of this study were to examine the expression of Scx-mScarlet and Col1a1-BFP in the embryo; quantify the density of Scx cells in the central region of the Achilles tendon of young and adult mice; and validate the tendon-specific dual reporter system by the in vitro stretch assay of adipose-derived mesenchymal stem cells (ADMSCs).
